ABSTRACT

Abstract Oil-in-water photoprotective nanoemulsions (NEs) were developed using Babassu (BBS) lipophilic extract, nonionic surfactants, and low concentrations of organic sunscreens by ultrasonic processing. BBS extract was chosen due to its suitable physicochemical properties (acidity index, peroxide index, refraction index, and relative density) and predominance of saturated fatty acids, identified by gas chromatography-mass spectrometry (GC-MS), which promote biological activities and high oxidative stability. NEs were characterized by mean droplet size, morphology, polydispersity index (PdI), pH, and organoleptic properties, and the physical stability of the NEs was evaluated for 120 days at room temperature. The sun protection factor (SPF) was determined, and the photostability and in vitro cytotoxicity assays were performed for NEs. All NEs remained stable for 120 days, with a droplet size <150 nm and a monomodal distribution profile. The pH values were compatible with the skin's pH. NE3 showed a spherical morphology, with a mean droplet size of 125.15 ± 0.16 nm and PdI of 0.145 ± 0.032. NE3 containing BBS extract and sunscreens presented an SPF of 35.5 ± 3.0, was photostable after 6 h of radiation and was non-cytotoxic to fibroblast cells. Thus, NE3 could be considered a promising formulation for developing synergic plant-extract sunscreen photoprotective products for the market

Resumen Introducción. El Zinc (Zn) es un oligoelemento esencial con gran importancia nutricional e indispensable para el crecimiento normal y la reproducción. Su deficiencia produce anormalidades fisiológicas y estructurales. Así mismo, ingerido en altas concentraciones produce efectos tóxicos, de allí la importancia de su determinación. El agua puede contribuir significativamente a la ingesta diaria de elementos trazas, entre ellos Zn. En el presente estudio se analizó el aporte de Zn en muestras de aguas de ríos y de red (de suministro público) de la provincia de San Luis y en agua mineral natural envasada, por medio de fluorescencia en fase sólida (FFS). Materiales y método.La determinación cuantitativa de Zn en muestras de agua y estándares se basó en la complejación del Zn utilizando una mezcla de o-fenantrolina y eosina a pH 7,5. Luego, una microzona de papel de filtro Blue Ribbon se impregna con la mezcla durante 1 minuto (n=6). Posteriormente, los papeles de filtro se secaron a temperatura ambiente y se colocan en una celda de cuarzo convencional adaptada para FFS. Los resultados obtenidos fueron comparados con la técnica de espectrometría de masas con plasma acoplado inductivamente. Resultados. Las muestras de agua de río analizadas presentaron valores por debajo del límite máximo de Zn recomendado por la Ley Nacional 24051, para la protección de la vida acuática. Las concentraciones de Zn en las muestras de agua de consumo analizadas (de red y envasadas) también estuvieron por debajo de los límites máximos permitidos por el Código Alimentario Argentino y la OMS. Por otro lado, se obtuvo buena concordancia entre las metodologías utilizadas. Conclusiones. El contenido de Zn de las muestras analizadas se encuentra por debajo de los valores máximos permitidos por los distintos organismos de regulación. Las muestras de agua analizadas no contribuyen a satisfacer la ingesta diaria recomendada y presentan valores considerablemente menores a los encontrados en otras regiones. Finalmente, el método aplicado mostró ser una alternativa rápida y sensible para la determinación de Zn en muestras de agua.


Abstract Introduction. Zinc (Zn) is an essential trace element with great nutritional importance and indispensable for normal growth and reproduction. Its deficiency produces physiological and structural abnormalities. Also, if ingested in high concentrations, it produces toxic effects; this is why its determination is very important. Water can contribute significantly to the daily intake of trace elements, including Zn. In this work, Zn concentration was analyzed in river and tap water samples of the province of San Luis and in bottled natural mineral water, by means of solid phase fluorescence (SPF). Materials and method. The quantitative determination of Zn in water samples and standards was based on the complexation of Zn using a mixture of o-phenanthroline and eosin at pH 7.5. Then, a microzone of Blue Ribbon filter paper was impregnated with the mixture for 1 minute (n=6). After this, the filter papers were dried at room temperature and placed in a conventional quartz cell adapted for SPF. The results obtained were compared with the Inductively Coupled Plasma - Mass Spectrometry (ICP-MS) technique. Results. The river water samples analyzed were found below the maximum limit of Zn recommended by National Law 24051 for the protection of aquatic life. The Zn concentrations in the analyzed drinking water samples (tap and bottled water) were also below the maximum limits allowed by the CAA and WHO. Furthermore, a good agreement was obtained between the methodologies used. Conclusions. The Zn content in analyzed samples was below the maximum values allowed by different regulatory organizations. The water samples analyzed do not contribute to satisfying the recommended daily intake and have considerably lower values than those found in other regions. Finally, the method applied proved to be a fast and sensitive alternative for the determination of Zn in water samples.

OBJECTIVE: To explore the effect of 1,25(OH)2D3 on the expression of microRNA-130b and transforming growth factor pi in kidney of diabetic nephropathy rats. METHODS: The study was approved by the Laboratory Animal Ethical Committee of Laboratory Animal Center of Xinjiang Medical University. ABSTRACT

The present study aimed at the evaluation of Passiflora coccinea (Aubl.) antioxidant and photo protective in vitro activities, looking forward to their application as antiaging or sunscreen agents in cosmetic formulations. Methanolic and glycolic leaf extracts were prepared by three methods: ultrasound assisted extraction (UAE, 30 min.), maceration at room temperature (72 h) and maceration at 30 ºC (72 h). The antioxidant activities of the extracts were measured by DPPH and ORAC-FL assays and they were incorporated into a cosmetic emulsion to have their sun protection factor (SPF) measured spectrophotometricaly. The antioxidant activity of the emulsions were measured by DPPH and ORAC as well. C-glycosyl-flavones were identified in the extracts by ESI-MS/MS, in comparision with standards. The UAE methanolic extract and the maceration at 30 ºC glycolic extract were submmited to HPLC-DAD analysis and isovitexin was quantifyed in both by a validated method. The methanolic extract antioxidant activity was independent of the extraction method, higher than reported for other species of Passiflora and detectable when incorporated into the emulsion formulation. Maceration at 30oC was the most suitable method for glycolic extraction and its antioxidant activity was lower than the value presented by the methanolic extracts. None of the extracts exhibited a SPF value. Isovitexin in the UAE methanolic extract was 12.67 times higher than the most active glycolic extract, aside of their similar chromatographic profiles. Although a SPF value was not detected, the results indicate that P. coccinea can be a potential new source of antioxidants for topical antiaging formulations.

ABSTRACT

Objective: Bemotrizinol (BEMT) is the most efficient broad-spectrum UV-absorber having a dual mechanism of action in absorbing and reflecting photons. The main objective of this work was to develop successful oil in water (o/w) nanoemulsion for improving the solubility of BEMT and its protective characteristics. Methods: Pseudo-ternary phase diagrams were constructed using labrafac PG and isopropyl myristate as oil phase, tween 80 as surfactant (S) and cremophor EL as cosurfactant (CoS) the ratio of S/CoS was determined according to highest percent of water incorporation to the system. Full factorial study design (24) using Design-Expert® software was adopted to study the effect of four independent variables namely: oil type, oil concentration, S/CoSmix (3:1) concentrations and BEMT concentration on the particle size and the in vitro release at 2 h (Q2h) of the prepared nanoemulsion formulae. Two systems each of eight formulae were developed and evaluated through droplet size analysis, zeta potential measurement, refractive index, in vitro drug release and according to the desirability value two formulae (F6 and F14) were used for further evaluations including in vitro sun protection factor (SPF), ex-vivo deposition by tape stripping technique, permeation test and photostability study. Results: Formula (F14) was chosen as the optimum formula having an in vitro SPF of 16.08±0.39, lowest permeation of 140±0.06 μg/cm2after six h and highest photostability (t90% = 168.02) after 120 min. Conclusion: Despite the poor solubility of bemotrizinol, it could be enhanced by novel drug delivery systems with good SPF value while maintaining its photostability.

Background: Use of sunscreens on the face is becoming popular, and patients with melasma are prescribed sunscreen for use on the face. Results of a few Western studies on the effect of sunscreen use on serum vitamin D concentration are not applicable to Indian conditions. Aims: To examine the effect of use of a high sun protection factor (SPF 50+, PA++++) sunscreen on face in patients with melasma on serum concentration of 25-hydroxyvitamin D. Methods: Forty-five Indian patients (Fitzpatrick skin types III and IV) with melasma were advised to use a sunscreen with SPF 50 + for 3 months, 43 (33 female, 10 male; age 32.9 ± 8 years) completed the study. Patients staying outdoor for <4 hours applied sunscreen once daily after bath. Patients staying outdoors for >4 hours reapplied sunscreen 4 hours after first application. Patients were provided a container to measure the amount of sunscreen for use, which was approximately equal to recommended thickness. Compliance was tested by weighing the used tubes and tubes in use during monthly visits. Serum concentration of 25-hydroxyvitamin D was tested before and after the study period. Results: Amount of sunscreen advised (100.5 ± 29.2 ml) and the actual amount used (96.6 ± 27.9 ml) were similar (P = 0.53, t-test). The difference between serum concentrations of 25-hydroxyvitamin D at the baseline (19.20 ± 9.06 ng/ml) and at 3 months (18.91 ± 8.39 ng/ml) was not significant (P = 0.87, paired t-test, 95% confidence interval of difference −3.33 to 3.92). No correlation was found between the amount of sunscreen used and the percentage change in serum 25-hydroxyvitamin D concentration at 3 months (rho = 0.099, P = 0.528, Spearman's rank correlation). Limitations: Longer duration of application and a larger sample size may detect minor differences in vitamin D concentration. Conclusion: Using a high SPF sunscreen on the face, along with physical photoprotection advice, in patients with melasma for 3 months does not influence serum 25-hydroxyvitamin D concentration in Indian conditions.

Objective@#To investigate the presence of hepatitis E virus (HEV) RNA and HEV antigen (HEV-Ag) and to evaluate the infectivity of HEV in urine through a SPF rabbit model of HEV infection.@*Methods@#Serum, fecal and urine samples collected from SPF rabbits with HEV infection were tested for viral and biochemical markers using real-time RT-PCR and ELISA. Liver and kidney biopsies were performed for observing histopathological changes and immunohistochemical staining. Rabbits were challenged with HEV isolated in urine samples to evaluate the infectivity.@*Results@#Rabbit R1# that was injected with rabbit HEV presented viremia, fecal shedding of HEV, high serum level of HEV-Ag, elevated aspartate transaminase (AST) and alanine transaminase (ALT) and typical symptoms of hepatitis. Urine samples of rabbit R1# continued to be positive for HEV RNA and HEV-Ag. Ratios of HEV-Ag to RNA in urine samples of rabbit R1# were significantly higher than those in serum and feces samples. The parameters quantified in routine urinalysis remained within the normal ranges in rabbit R1#. However, pathological changes and the presence of HEV-Ag were observed in kidney tissues. Furthermore, serum and fecal samples that were collected from one of the two rabbits injected with rabbit R1# urine-derived HEV were HEV positive and the virus strains isolated form feces remained infective to rabbits.@*Conclusion@#HEV infection may result in kidney injury and the urine may pose a risk of transmission. HEV-Ag detection in urine may be valuable for the diagnosis of ongoing HEV infection.

ABSTRACT

Objective To study the effect of age and sex on physiological and biochemical indexes of SJ5-SPF chickens at the age of different weeks. Methods Blood biochemical indexes of SJ5-SPF chickens at the age of different weeks were measured using an automatic blood analyzer, including 19 items: ALT, ALP, GGT, AST, LDH, TP, GLB, ALB, TB, DB, IBIL, GLU, TG, TCH, BUN, CRE, K+, Na+, and Ca2+. At the same time, the body temperature (T), respiratory frequency (R), heart rate (HR), diastolic pressure (DBP) and systolic pressure (SBP) of the SJ5-SPF chickens were measured using a RM6240C multi-channel physiological signal acquisition and processing system. Results (1) Among the physiological parameters, the body temperature, respiratory rate, heart rate, diastolic blood pressure and systolic blood pressure showed significant differences between the 4-week old chickens and the chickens at ages of 20, 25, and 25 weeks (P < 0. 05), but there were significant differences between the males and females at the same weeks of age except body temperature. The diastolic blood pressure was only significantly different in the 40-week old male and female chickens (P < 0. 01). The heart rates showed significant differences between the male and female chickens at 4, 20 and 25 weeks of age (P < 0. 01). The respiratory rate and systolic blood pressure in the 4-week old male and female chickens were significantly different (P < 0. 05). (2) The 19 blood biochemical indexes showed significant differences between the male and female chickens: the GGT, ALT, AST, TB, DB, IBIL, GLU, BUN and Na+at an week-age (P< 0. 05), the LDH, TP, GLB, ALB and CRE at two different week-ages (P< 0. 05), the ALP, TG and Ca2+at three different week-ages (P< 0. 05), and the TCH and K+at four different week-ages (P< 0. 05). Conclusions The above results can provide a scientific basis for diagnosis of SJ5-SPF chicken disease, animal quarantine and other related research.

Objective To analyze the changes and gender differences of organ coefficients, intestinal length and body size of SJ5-SPF chickens at different weeks of age,and to provide experimental data for the sex selection of laboratory chickens. Methods The body weight,the weight of 15 main organs,and length of 5 main segments of intestine and the 6 main parameters of body size of SJ5-SPF chickens at the age of 4,20,25 and 40 weeks were measured,respectively,and the organ coefficients were calculated. In addition, the gender differences in body weights, organ coefficients, intestinal lengths and body sizes between the female and male SJ5-SPF chickens at the four different weeks of age were compared, respectively. Results There were significant differences in body weights between the male and female SJ5-SPF chickens at each of the four different weeks of age(P < 0.01). The organ coefficients varied between females and males to different extents at different weeks of age. As for the intestinal length,there were no significant differences in the length of jejunum and ileum as well as rectum between the male and female chickens, while the length of duodenum, left cecum and right cecum showed gender differences at some of the four different weeks of age. During the determination of body size, there were gender differences in body length,shank length,pelvic width,chest depth and chest width at two or three of the four different weeks of age. Conclusions The organ coefficients, intestinal length and body size of SJ5-SPF chickens are affected by both their age and gender.

ABSTRACT

Filtros orgânicos são amplamente utilizados em formulações fotoprotetoras, com habilidade de absorver radiações ultravioleta (UV). Contudo, parte destes compostos possuem limitações, como: fotoinstabilidade, permeação cutânea e fotossensibilização e entre outros. Este trabalho envolveu a síntese de matriz mesoporosa do tipo SBA-15, encapsulação/incorporação de ρ-metoxicinamato de octila (MCO), benzofenona-3 (BZF-3) e avobenzona (AVO) na SBA-15 para aplicação em formulações fotoprotetoras. Fez-se a determinação da eficácia in vitro dos filtros encapsulados/incorporados combinados a ingrediente cosmético; o preparo de bastão fotoprotetor e determinação eficácia estimada; a avaliação do potencial de irritação ocular dos bastões por HET-CAM - Hen's Egg Test - Chorioallantoic Membrane, e a avaliação da permeação/retenção cutânea e perfil de biodistribuição dos filtros. Para a caracterização dos materiais foram empregadas técnicas físico-químicas e analíticas. As medidas de adsorção/dessorção de N2 mostrou que as amostras dos filtros solares encapsulados/incorporados apresentaram diminuição na área superficial e volume de poro (V), indicando que os filtros solares foram encapsulados/incorporados na superfície e nos poros da SBA-15. Os resultados de Espalhamento de raios X a baixo ângulo evidenciaram que os filtros solares não afetaram a estrutura hexagonal da SBA-15. Por TG/DTG e análise elementar foi possível determinar a quantidade de filtros solares na superfície e nos mesoporos da SBA-15. Enquanto, as curvas DSC e DTA revelaram aumento na estabilidade térmica da AVO e BZF-3, quando encapsulados/incorporados. Os resultados da eficácia fotoprotetora in vitro mostraram que a combinação dos três filtros solares encapsulados/incorporados na SBA-15 promoveram aumento de 52% no fator proteção solar (FPS), enquanto que, na formulação fotoprotetora contendo os três filtros encapsulados/incorporados, o aumento foi de 94%. O ensaio de HET-CAM evidenciou que os bastões contendo SBA-15 e os filtros encapsulados/incorporados não foram irritantes. O ensaio de permeação/retenção cutânea mostrou que o processo de encapsulação/incorporação da BZF-3 promoveu diminuição de sua permeação em todos os tempos de exposição. As quantidades permeadas de AVO e MCO ficaram abaixo do limite de quantificação nos tempos 6 e 12 h, no entanto, no tempo de 24 h foi possível quantificá-los. As quantidades dos filtros solares retidas na pele a partir da formulação contendo os filtros solares encapsulados/incorporados na SBA-15 (F4) foram menores (tempos 6 e 12 h) em comparação à formulação contendo os filtros solares não encapsulados (F3). A investigação da biodistribuição dos filtros solares mostrou que a retenção total na pele, como na derme, foi menor na formulação F4 em comparação à F3. O estudo comparativo entre pele suína e a pele humana mostrou que as quantidades de BZF-3 e AVO permeadas e retidas na pele suína foram superiores do que em relação à humana para ambas as formulações (F4 e FR). Pela técnica de biodistribuição, foi possível determinar que os filtros solares oriundos das formulações F3 e referência (FR) apresentaram maior retenção destes compostos na derme do que em outras camadas da pele. Contudo, observou-se que os filtros encapsulados apresentaram taxa reduzida de retenção na derme


Organic Filters are chemical compounds widely used in sunscreens formulations with the ability to absorb ultraviolet radiation (UV). Despite the effectiveness of these compounds in UV radiation protection, disadvantages related to their photo instability, potential skin permeation and photo sensibility pose significant challenges for improving these products. The aim of this work was to synthesize mesoporous matrix SBA-15, encapsulation/entrapping of octyl methoxycinnamate (OMC), benzophenone-3 (BZF-3) and avobenzone (AVO) into SBA-15 for application in photoprotective formulations. It was accessed in vitro photoprotection efficacy and in vitro photostability assay of encapsulated/entrapped UV filters combined with cosmetic ingredient and photoprotective stick formulations; evaluation of the ocular irritation potential of photoprotective stick formulations by in vitro method HET-CAM - Hen's Egg Test - Chorioallantoic Membrane; evaluation the skin permeation/deposition and biodistribution profile of photoprotective stick formulations. The decrease in the surface area and in mesoporous volume (V) observed in the nitrogen adsorption desorption isotherms of encapsulated/entrapped samples indicated that UV filters were efficiently encapsulated/entrapped into SBA-15. Additionally, SAXS results showed that UV filters did not affected the hexagonal structure of the mesoporous material and that these compounds filled the SBA-15 pores. TG/DTG and elemental analysis were efficient tools to confirm the presence and the quantity of UV filters into SBA-15. DTA and DSC curves of encapsulated/entrapped materials showed that the thermal stability of AVO and BZF-3 were increased. On the other hand, DSC curves of encapsulated/entrapped materials demonstrated that thermal stability of OMC was not increase. The in vitro photoprotective efficacy results demonstrated that the combination of the three sunscreens encapsulated/entrapped into SBA-15 increased 52.0% the SPF values, while the stick formulation containing the UV filters encapsulated/entrapped, the increase was 94.0%. Delivery experiments using porcine skin demonstrated that the encapsulation/entrapping process of UV filters resulted the decreased of BZF-3 permeation and deposition in skin (6 and 12 hours). The cutaneous biodistribution profile of UV filters showed that the deposition of these compounds from encapsulated/entrapped stick formulation (F4) was significantly lower than that from UV filters stick formulations (F3) in the total slices of the skin and dermis. The comparative study between porcine skin and human skin demonstrated that the amounts of BZF-3 and AVO permeated and deposited in porcine skin were higher than in human skin for both formulations (F4 and FR - reference formulation). By the biodistribution technique it was possible to determine that the UV filters from the formulations F3 and FR presented higher retention of these compounds in the dermis than in other layers of the skin. On the other hand, it was observed that the encapsulated UV filters presented low retention rate into dermis

Objective The aim of this study was to investigate the polymorphism of SLA class II genes in Canadian SPF Yorkshire and Landrace pigs.Methods Blood samples were obtained from 15 SPF Yorkshire and 22 Landrace pigs for isolation of peripheral blood mononuclear cells respectively, and the DQB1, DRB1 and DQA genes were amplified by PCR after reverse transcription.SLA class II genes were obtained by analyzing the direct and cloning result.The polymorphism of alleles was analyzed using the DNAsp 5.0 software.Results A total of 25 alleles were identified at three genes, including eight DQB1, ten DRB1 and seven DQA, and three alleles were submitted the complete sequences for the first time.The official allele names were assigned as SLA-DQB1*0212 (KU754590), SLA-DQB1*0203 (KU754591) and DRB1*06:07(KU754601) by the SLA Nomenclature Committee.Three novel DQA alleles were discovered.Five of the 15 amino acids, one of the 16 amino acids and 11 of the 19 amino acids, which bind processing antigens, showed well conserved among the alleles of DQB1, DRB1 and DQA genes in the SPF Yorkshire and Landrace pigs, respectively.Neighbor-joining tree showed that the three genes were divided into two clusters, respectively.There was a close relationship between SPF Yorkshire and Landrace pigs and foreign Yucatan miniature pigs, and it showed no obvious genetic distance with other pigs.Conclusions A total of 25 SLA class II alleles have been identified successfully in this study, and there are more abundant polymorphism for them.There is a widely distribution for SLA class II alleles identified in this study in other pig breeds.It is critical for the eventual future use of SPF Yorkshire and Landrace pigs as classical laboratory animal models.

Objective To determine the reproductive physiology and blood physiological and biochemical characteristics of SPF Yorkshire and Landrace swine.Methods Ten reproductive physiology parameters,19 blood physiological parameters and 18 blood biochemical parameters in SPF Yorkshire and Landrace swine were measured using conventional methods and the differences between population,between age groups and between both sexes were analyzed.Results There were no significant differences(P>0.05) in reproductive physiology parameters and most blood physiological and biochemical parameters of the SPF Yorkshire and Landrace swine.A few of parameters,such as blood physiological indices GRAN,HGB,RDW,PLT,PCT,and blood biochemical indices ALKP,CHOL,TBIL,BUN,showed significant difference(P<0.05) between populations,between age groups and between both sexes,however,the values of difference were rather small,deviated from the normal range.Conclusion The physiological and biochemical characteristics of SPF Yorkshire and Landrace swine are basically stable and there is no significant difference compared with other laboratory miniature pigs.This study will provide valuable basic data for raising velvet yield,establishment of animal models and evaluating the genetic quality of closed colony.

A exposição crônica à radiação solar pode contribuir para o aparecimento do câncer de pele, sendo o uso de fotoprotetores um fator primordial na prevenção desses efeitos deletérios. Atualmente, substâncias bioativas tais como a rutina têm sido foco de interesse da comunidade científica graças às suas propriedades fotoprotetoras e antioxidantes, que podem promover aumento dos valores de FPS, além de conferir características multifuncionais às formulações. Achados in vitro recentes indicam que a rutina, quando incorporada em emulsões fotoprotetoras óleo em água, promove aumento da atividade antioxidante e aumento do FPS. No entanto, a realização de estudos clínicos é fundamental para confirmar e quantificar esses resultados, já que a metodologia in vitro possui baixa repetibilidade e ausência de correlação com ensaios in vivo, principalmente quando as formulações analisadas apresentam substâncias antioxidantes em sua composição. O objetivo deste estudo foi avaliar pela primeira vez a atividade da rutina frente ao FPS e sua segurança clínica através da comparação de formulações fotoprotetoras contendo rutina 0.1% (w/w), avobenzona 3.0% (w/w) e octil dimetil PABA 8.0% (w/w) com uma preparação similar sem o composto bioativo. Adicionalmente, hidratação cutânea, FPS in vitro e atividade antioxidante da rutina em associação com outros filtros foram investigados. O perfil de segurança das formulações qualificou as fórmulas para os testes de eficácia clínica. O teste de DPPH confirmou a capacidade antioxidante da rutina, demonstrando cerca de 40% de aumento na capacidade de sequestro de radicais livres na presença do composto bioativo. A rutina em combinação com os filtros UV aumentou o FPS clínico de 7.30 ± 0.60 para 12.37 ± 1.13, o que representa cerca de 70% de aumento. Os resultados encontrados provam que a rutina em combinação com outros filtros pode aumentar significativamente o valor do FPS e que a mesma é segura para uso clínico


Unprotected chronic exposure to solar radiation can contribute to premature skin cancer and sunscreens are a key factor to avoid those detrimental effects. Currently, there is a growing interest in the photoprotector and antioxidant potential of bioactive substances, such as rutin, that could help to increase the SPF value and add multifunctional characteristics to the formulations. Recent in vitro findings indicated that rutin, when incorporated in oil-in-water photoprotective emulsions can provide antioxidant activity and SPF increase. However, clinical studies are fundamental to determine this activity duo to in vitro methodology lack of repeatability and correlation between the in vivo data, especially when the analyzed formulas contain antioxidant substances. The aim of this study was to evaluate for the first time to date the rutin in vivo SPF and clinical safety by comparing sunscreens formulations containing rutin 0.1% (w/w), butyl methoxydibenzoylmethane 3.0% (w/w) and octyl dimethylPABA 8.0% (w/w) with a similar bioactive-free preparation. Additionally, skin hydration, in vitro SPF and in vitro antioxidant activity of rutin, in association with the UV filters were investigated. The safety profile of the formulations under sun-exposed skin conditions qualified the formulas for clinical efficacy assays. DPPH test confirmed rutin antioxidant properties, demonstrating about 40% increase in radical scavenging potential when the bioactive compound was present. Rutin in combination with the UV filters increased the clinical SPF from 7.30 ± 0.60 to 12.37 ± 1.13, representing about 70% growth in the SPF value. The results obtained proved that rutin in combination with UV filters can improve the SPF value significantly and is safe for clinical use

ABSTRACT

Avobenzone is UVA sunscreen active substances, which are unstable when exposed to UV radiation, especially UVA. Theaim of this study is to determine the effectiveness and photo stability avobenzone when combined with ethyl ascorbic acid and alpha tocopherol acetate with various concentrations of ethyl ascorbic acid and alpha tocopherol by in vitro using spectrophotometric method. The photostability study of the eight variations of formula showed that the formula3, 4, 5, 6, 7 and 8 had significant differences with Formula 1 (p<0,05) and the best photostability represented by the formula 4 (avobenzone 2%, ethyl ascorbic acid2%) with avobenzone concentrations decrease by 11,82% for 15 hour using UVA lamp irradiation 4,7 mW/cm2. In effectiveness of SPF (Sun Protection Factor) value determination showed moderate protection category (SPF ±6). The percent transmission of erythema and pigmentation test showed that cream had effectiveness as sunscreen by showing the category as sun block on the area pigmentation and not to erythema areas at concentration 1000 μg/ml.

Objective To investigate the positive and suspicious rates of BV, SRV, SIV and STLV-1 antibody of the cynomolgus monkeys introduced from Laos, dynamic monitoring of virus antibody during the establishment of SPF cyno-molgus monkey population, and then comparing the positive rate of virus antibody in the baby monkeys between the common with SPF populations.Methods Continuous monitoring four kinds of virus in the monkeys was carried out using special reagent kits and analyzed the data.Results In the introduced cynomolgus monkeys (n=1998), the positive rate of BV antibody was 52.35%, suspicious rate of BV antibody was 8.31%, and negative rate of BV antibody was only 39.34%, the positive rates of SRV and STLV-1 antibody were 7.45%and 8.56%, respectively, and positive or suspicious SIV anti-body in the cynomolgus monkeys was not detected.After screening and establishment of the SPF population, the positive rates of BV, SRV and STLV-1 antibody were 5.24%, 1.01%and 0.4%, respectively, in the year 2010.Then after five years of screening and elimination, by the end of 2014, the positive rates of BV, SRV and STLV-1 antibodies were 0.82%, 0.27%and 0.27%, respectively, and positive or suspicious SIV antibody in the cynomolgus monkeys was not de-tected.The BV antibody positive rate of the baby monkeys was 9.71%and the suspicious rate was 1.85%in the common population, but only 0.22%of the baby monkeys in the SPF population.Conclusions Continuous monitoring virus anti-bodies and eliminating positive and suspicious animals have important significance in the establishment of a SPF cynomolgus monkey population.

Objective To accurately determine the biological characteristics , especially the disease-related indexes of SPF rabbits , and their gender differences .Methods A total of thirty 70-80 day old SPF New Zealand rabbits (male :female=1:1) were fed for a week, and then we weighed the body weight and main organs , determined the blood physiological and biochemical indexes , blood gas and arterial pressure , and measured the ventricular pressure by thoracotomy under respiratory support .Results Comparison of the male and female SPF New Zealand rabbits showed that there were significant differences in the mass of thyroid , adrenal gland, and liver (P<0.05 or P<0.01); the brain, pituitary gland, thyroid gland organ coefficients ( P<0.05 or P<0.01); the thyroid/brain, adrenal/brain, and liver/brain ratios (P<0.05 or P<0.01);the mean erythrocyte volume and mean hemoglobin content (P<0.05 or P<0.01);and in glutamyl transpetidase and amylase ( P<0.05 or P<0.01 ) , but no significant differences in blood gas analysis ,heart rate and carotid artery systolic and diastolic blood pressure , left ventricular systolic and diastolic blood pressure , and right ventricular systolic and diastolic blood pressure .Conclusions The gender has an impact on organ weight and blood physiological and biochemical indexes in New Zealand rabbits , but no significant influence on blood gas , blood pressure , heart rate and ventricular pressure in the rabbits .

Se evalúa la influencia de la altitud sobre el Índice de Protección Natural (IPN) y el Factor de Protección Solar requerido (SPF) comparando niños de Mucuchies, Mérida, Venezuela (3100 m.s.n.m.) y Palmarito, Mérida, Venezuela (5 m.s.n.m.), utilizando el equipo Sun Protection Diagnostic SP 37®. Según el análisis de los resultados a través de estadísticas descriptivas, se evidencia que el IPN y el SPF son variables dependientes de la altitud de la localidad en que se encuentran los niños, ya que en las localidades más altas los rayos solares inciden de forma más directa que en las más bajas.


It is evaluated the effect of altitude on Natural Protection Index (IPN) and the required Sun Protection Factor (SPF) comparing children Mucuchies, Merida, Venezuela (3100 m) and Palmarito, Merida, Venezuela (5 m) using Sun Protection Diagnostic SP37® equipment. According to the analysis of results through descriptive statistics, it appears that the IPN and the SPF are variable dependent on the altitude of the locality in which children are, as in the most high solar rays more directly than in the lowest.

ABSTRACT

The aim of the present study was to determine the ultraviolet (UV) absorption properties of aqueous herbal extracts of some commonly found vegetable sources by determining the sun protection factor (SPF) number. The in vitro SPF number is determined according to the spectrophotometric method described by Mansur et al. Aqueous herbal extracts were prepared and after dilution with alcoholic solutions the absorbance were recorded between 290-320 nm using UV-vis spectrophotometry. It was observed that all of the tested herbals showed some UV protection capabilities with aqueous coconut extract showing the highest SPF number of 7.38 while watermelon showed the lowest SPF number of 0.97.
